Description: NADPH-diaphorase substrate that competitively inhibits NOS (nitric oxide synthase). Well-known scavenger of superoxide anions. Dye that is used for detection of alkaline phosphatase in combination with 5-bromo-4-chloro-3-indoxyl phosphate (BCIP). This substrate system produces an insoluble NBT diformazan end product that is blue in color and can be observed visually. When used with BCIP, it is suitable for detection of alkaline phosphatase in western blots, for immunohistological staining procedures and for colorimetric indication of bacterial infection in blood samples. Used as a redox indicator for enzymatic reactions including dehydrogenases, threonine deaminase, glucose-6-phosphate dehydrogenase, phosphofructokinase on polyacrylamide gels, oxidases on polyacrylamide gels and pentose shunt dehydrogenses. The NBT/BCIP reaction is also used for colorimetric/spectrophotometric activity assays of oxidoreductases. One application is in activity stains in gel electrophoresis, such as with the mitochondrial electron transport chain complexes.

Description: UCP2 (uncoupling protein 2) is a member of the mitochondrial carrier protein super family located in the mitochondrial inner membrane. The UCPs differ greatly in tissue distribution: UCP1 is expressed only in brown adipose; UCP3 preferentially in skeletal muscle, brown adipose and heart; UCP4 and UCP5 mainly in the nervous system (18167556). In contract, UCP2 has a broad distribution and is implicated in a wide range of pathophysiological processes, including diabetes and cancer. This antibody detect a band around 28-33 kDa corresponding to UCP2 protein in mouse liver, skeletal muscle and pancreas. Some upper and lower bands also were observed due to the dimer formation and degradation of the UCP2 protein (20103532). This antibody was raised against the full-length UCP2 protein and may cross-react with UCP3. It is notable that UCP2 is known to have a very short half-life less than 1 hour (17240372). It is strongly recommended that fresh-made lyate be used for western blot assays.

Description: Protein tags are protein or peptide sequences located either on the C- or N- terminal of the target protein, which facilitates one or several of the following characteristics: solubility, detection, purification, localization and expression. Maltose binding protein (MBP) is the 370 amino acid product of the E.coli mal E gene. MBP is a useful affinity tag that can increase the expression level and solubility of the resulting tagged protein. The MBP tag also promotes proper folding of the attached protein. Plasmid vectors have been constructed utilizing the MBP domain that allow the synthesis of high levels of MBP-fusion proteins that can be purified in a one step procedure by affinity chromatography cross linked amylose resin. Once bound to amylose, the MBP protein can then be separated from the target protein by cleavage by coagulation Factor Xa at a specific four residue site. Alternatively, the intact fusion protein can be specifically eluted from the resin by the addition of excess free maltose. Subsequent to elution, MBP fusion protein can be visualized either by western blot analysis or immunoprecipitation using antibodies specific for the MBP-tag. This antibody recognizes MBP (Maltose binding protein) TAG in some expression systems.

Description: The protein named TrkB (also named Neurotrophic tyrosine kinase receptor type 2 (NTRK2), GP145-TrkB or Tropomyosin-related kinase B is a receptor tyrosine kinase involved in the development and the maturation of the central and the peripheral nervous systems and is important in the regulation of neuron survival, proliferation, migration, differentiation, and synapse formation and plasticity. TrkB may also play a role in neutrophin-dependent calcium signaling in glial cells and mediate communication between neurons and glia. TrkB is the primary receptor for BDNF (brain-derived neurotrophic factor. TrkB also binds NT4 and NT3 but less efficiently (Reference: www.uniprot.org). Antibody reacts with human and mouse, other species not yet tested but it is predicted to react with rat and chicken TrkB-pS478 based on amino acid homology at the site. Antibody has been shown to be specific for TrkB phosphorylated on serine 478 by phospho-peptide absorption dot blots, and on cell lysates from cell lines induced with retinoic acid and BDNF. Antibody detects a clear band in retinoic acid (RA) and BDNF-treated NSC34 cell lysates at 140 kDa only, indicating that the phosphorylated TrkB receptor is being detected. Additional non-specific bands at lower molecular weight are observed in both lysates with the antibody and these bands have not been characterized.
