Description: T cells require a signal induced by the engagement of the T cell receptor and a “costimulatory” signal(s) through distinct T cell surface molecules for optimal T cell activation and tolerance. CD273 is one of two ligands for programmed death-1 (PD-1; CD279), a member of the CD28 family of immunoreceptors. The other identified ligand is PD-L1. CD273 is broadly expressed and also up regulated in a variety of tumor cell lines. On previously activated T cells, CD273 interaction with PD-1 inhibits TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. CD273 has a costimulatory function on resting T cells activated with suboptimal TCR signals.

Description: Interleukin-6 (IL-6) is a multifunctional factor, which exerts its activities through binding to a highaffinity receptor complex consisting of two membrane glycoproteins: an 80 kDa component receptor that binds IL-6 with low affinity (IL-6Ralpha) and a signal transducing component of 130 kDa (gp130) that does not bind IL-6 by itself, but is required for highaffinity binding of IL-6 by the complex. A soluble form of the IL-6Ralpha has been found in the urine of healthy adult humans.

Description: CD200, also known as OX-2, is a 45 kDa transmembrane immunoregulatory protein that belongs to the immunoglobulin superfamily. CD200 is widely but not ubiquitously expressed. Its receptor (CD200R) is restricted primarily to mast cells, basophils, macrophages, and dendritic cells, which suggests myeloid cell regulation as the major function of CD200. CD200 knockout mice are characterized by increased macrophage number and activation and are predisposed to autoimmune disorders. In T cells, CD200 functions as a costimulatory molecule independent of the CD28 pathway. Several viruses encode CD200 homologs which are expressed on infected cells during the lytic phase. Like CD200 itself, viral CD200 homologs also suppress myeloid cell activity, enabling increased viral propagation.

Description: Programmed death ligand 1 (PD-L1, B7-H1 or CD274) is a member of the growing B7 family of immune proteins that provide signals for both stimulating and inhibiting T cell activation. CD274 has been identified as one of two ligands for programmed death 1 (PD-1), a member of the CD28 family of immunoreceptors. CD274 is widely expressed in several organs such as heart, skeletal muscle, placenta and lung, and in lower amounts in thymus, spleen, kidney and liver. CD274 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. CD274 expression is also induced in dendritic cells and keratinocytes after IFN-gamma stimulation. CD274 expression is also upregulated in a variety of tumor cell lines. Interaction of CD274 with PD-1 results in inhibition of TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. The CD274 - PD-1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.

Description: FGF-19 is involved in the suppression of bile acid biosynthesis through down-regulation of CYP7A1 expression, following positive regulation of the JNK and ERK1/2 cascades. Stimulates glucose uptake in adipocytes. FGF-19 plays an important role in brain development during embryogenesis. FGF-19 binds only to FGF receptor 4 (FGFR4). Activity requires the presence of KLB.

Description: Sirtuin-2 is a NAD-dependent protein deacetylase, which deacetylates the 'Lys-40' of a-tubulin. It is involved in the control of mitotic exit in the cell cycle. Furthermore, a variety of age-associated diseases such as type 2 diabetes, obesity, osteoporosis, and Alzheimer's disease are associated with sirtuins. Selective inhibition of Sirtuin-2 achieves neuroprotection and identifies Sirtuin-2 to be a potential therapeutic avenue in Huntington's and Parkinson's disease.

Description: Interleukin-33 (IL-33; HF-NEV; IL-1F11), a member of the IL-1 family of cytokines, is expressed by many cell types following pro-inflammatory stimulation and is thought to be released on cell lysis. The 30kDa human IL33 is converted by CASP1 to a 18kDa protein. IL33 binds to and signals through ST2 (IL1R1) and its stimulation recruits MYD88, IRAK, IRAK4, and TRAF6, followed by phosphorylation of ERK1 (MAPK3)/ERK2 (MAPK1), p38 (MAPK14), and JNK. The ability of IL-33 to target numerous immune cell types, like Th2-like cells, mast cells, and B1 cells, and to induce cytokine and chemokine production underlines its potential in influencing the outcome of a wide range of diseases, such as arthritis, asthma, atopic allergy & anaphylaxis, cardiovascular disease/atherosclerosis, nervous system diseases, and sepsis.

Description: Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. CD45 plays an essential role in lymphocyte activation.
