Description: Adipose tissue of an organism plays a major role in regulating physiologic and pathologic processes such as metabolism and immunity by producing and secreting a variety of bioactive molecules termed adipokines. One highly conserved family of adipokines is adiponectin/ACRP30 and its structural and functional paralogs, the C1q/tumor necrosis factor-alpha-related proteins (CTRPs) 1-7. Unlike adiponectin, which is expressed exclusively by differentiated adipocytes, the CTRPs are expressed in a wide variety of tissues. These proteins are thought to act mainly on liver and muscle tissue to control glucose and lipid metabolism. An analysis of the crystal structure of adiponectin revealed a structural and evolutionary link between TNF and C1q-containing proteins, suggesting that these proteins arose from a common ancestral innate immunity gene. Of the CTRPs, CTRP2 is most similar structurally and functionally to adiponectin. Recombinant CTRP2 rapidly activated AMPK and MAPK in cultured C2C12 cells, leading to increased glycogen accumulation and fatty acid oxidation.

Description: ATP-binding cassette (ABC) transporters are an evolutionarily conserved family that use ATP hydrolysis to catalyze the transport of various molecules across cell membranes. They are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). ABCA7, a member of the ABC1 subfamily, is a full-size ABC transporter consisting of two sets of the multiple membrane-spanning domains plus the Walker motifs for the ATP interaction. Description: Bst2 was first identified as a novel terminal B-cell-restricted antigen termed HM1.24 that is expressed on certain bone marrow cells as well as other tissues. Description: Bub1 was initially identified as the mammalian homolog to the yeast protein through a cDNA screen. Bub1 encodes a kinase involved in the formation of the spindle checkpoint, an important mechanism that ensures high fidelity mitotic chromosome segregation. It is thought to be required for assembly of a functional inner centromere, sister chromatid cohesion via targeting of the Shugoshim protein and metaphase congression. Bub1 functions by phosphorylating cdc20, a member of the mitotic checkpoint complex and activating the spindle checkpoint. A related protein kinase Bub3 interacts with Bub1 and targets it to kinetochores prior to chromosome alignment. Mutations in bub1 have been associated with aneuploidy and several forms of cancer.
