Description: CARMA proteins belong to the membrane-associated guanylate kinase-like (MAGUK) family of proteins that can function as molecular scaffolds that assist assembly of signal transduction molecules. CARMA1, CARMA2, and CARMA3 share high degrees of sequence and functional homology, but their tissue-specific distribution suggests that they serve distinct biological functions in different cell types. As with CARMA1, the CARD domain of CARMA3 has been shown to specifically interact with BCL10, a protein known to function as a positive regulator of cell apoptosis and NF-kB activation. When expressed in cells, this protein binds to BCL10 and activates NF-kB Recent experiments have shown that CARMA3 is required for EGF-induced NF-kB activation and contributes to tumor growth in vivo, suggesting that CARMA3 may serve as a new therapeutic target for the treatment of EGFR-driven tumors.

As with CARMA1, the CARD domain of CARMA2 has been shown to specifically interact with BCL10, a protein known to function as a positive regulator of cell apoptosis and NF-kB activation. When expressed in cells, this protein activated NF-kB and induced the phosphorylation of BCL10 Alternative splicing of CARMA2 results in isoforms that possess differential effects on NF-kB activation and endoplasmic reticulum stress-induced cell death.

Description: Calcium/calmodulin-dependent serine protein kinase (CASK) is a conserved multi-domain scaffolding protein involved in brain development, synapse formation, and establishment of cell polarity. CASKINs (CASK interacting proteins) interact with CASK and is thought to play a role in CASK function, specifically by coupling CASK to distinct downstream effectors. CASKIN1 is a multidomain protein containing six N-terminal ankyrin repeats, one SH3 domain, and two sterile alpha motif domains followed by a long proline-rich sequence and a short conserved C-terminal domain. CASKIN1 coassembles with CASK on the cytoplasmic tail of neurexin 1, suggesting that CASKIN and CASK coat the cytoplasmic tails of neurexins and other cell-surface proteins.

Description: Three distinct signaling pathways lead to programmed cell death (apoptosis). The death receptor and mitochondrion pathways are the mains, in which the key apoptotic proteases capase-8 and caspase-9, respectively, are involved. The endoplasmic reticulum (ER) stress is the third apoptotic pathway and caspase-12 is involved. Caspase-12 is localized to the ER but not to cytoplasm or mitochondrion. Caspase-12 is activated by ER stress, including disruption of ER calcium homeostasis, and mediates ER stress-induced apoptosis. Caspase-12 is co-localized to the ER with several proteins that are involved in Alzheimer's disease including gamma-secretase presenilin and beta-amyloid precursor protein (APP). Caspase-12 mediates cytotoxicity induced by amyloid-beta. Caspase-12 is ubiquitously expressed in mouse tissues.

Description: CDKN3 is a member of the dual specificity protein phosphatase family that acts as a cyclin-dependent kinase inhibitor (1). CDKN3 has been shown to interact with and dephosphorylate specifically the CDK2 kinase thereby preventing it’s activation. CDKN3 has been reported to be deleted, mutated, or overexpressed in several kinds of cancers. Increased expression of CDKN3 leads to increased levels of kinase-associated phosphatase activity that inhibits the G(1)/S transition of the cell cycle by dephosphorylating the cyclin-dependent kinases (2). CDKN3 Protein is ideal for investigators involved in Signaling Proteins, Cell Cycle Proteins, Cancer, Cell Cycle, and Ser/Thr Kinases research.

Description: Accurate chromosome segregation during mitosis requires a kinetochore to assemble correctly at the centromere of each chromatid and form a dynamic interface with the microtubules of the mitotic spindle. The kinetochore assembly includes the multisubunit CENP-H/I complex which can be divided into three functional classes based on phenotype analysis. One of these classes is referred to as the CENP-O class of proteins of which CENPO is the exemplar. These proteins form a stable complex with each other and are required for proper kinetochore function and for recovery from mitotic spindle damage. CENPO has also been shown to be an auto-antigen in a small population of anti-centromere antibodies- (ACA-) positive patients with sclerodoma.

Description: CHCHD7 is a newly identified member of a multifamily of proteins containing a conserved (coiled coil 1)-(helix 1)-(coiled coil 2)-(helix 2) domain. Northern blot analysis revealed that the gene is ubiquitously expressed. Its biological function is unknown and the gene has hitherto not been associated with neoplasia. CHCHD7 and PLAG1 are located head-to-head about 500 bp apart in 8q12. A chromosomal aberration involving CHCHD7 is found in salivary gland pleiomorphic adenomas, the most common benign epithelial tumors of the salivary gland.

Description: The fragmentation of nuclear DNA is a hallmark of apoptotic cell death. The activities of caspase and nuclease are involved in the DNA fragmentation. Caspase-activated deoxyribonuclease (CAD), also termed DNA fragmentation factor (DFF40), is one such nuclease, and is capable of inducing DNA fragmentation and chromatin condensation after cleavage by caspase-3 of its inhibitor ICAD/DFF45. Caspase and CAD independent DNA fragmentation also exists. Recent studies demonstrated that another nuclease, endonuclease G (EndoG), is specifically activated by apoptotic stimuli and is able to induce nucleosomal fragmentation of DNA independently of caspase and DFF/CAD. EndoG is a mitochondrion-specific nuclease that translocates to the nucleus and cleaves chromatin DNA during apoptosis. The homologue of mammalian EndoG is the first mitochondrial protein identified to be involved in apoptosis in C. elegans . EndoG also cleaves DNA in vitro.

Description: EZH2 is a histone-lysine methyltransferase which methylates ‘Lys-9’ and ‘Lys-27’ of histone H3, leading to transcriptional repression. It is a member of the polycomb group (PcG) family which form multimeric protein complexes and are involved in maintaining the transcriptional repressive state of genes over successive cell generations. The EZH2 activity is dependent on the association with other components of the PRC2 complex (EED, EZH2, SUZ12/JJAZ1, RBBP4 and RBBP7). EZH2 may play a role in the hematopoietic and internal nervous systems. Over-expression of EZH2 is observed during advanced stages of prostate cancer and breast cancer. Anti-EZH2 Antibody is ideal for research in Epigenetics, Gene Expression, Cell Biology and Cancer.

Description: Adipose tissue of an organism plays a major role in regulating physiologic and pathologic processes such as metabolism and immunity by producing and secreting a variety of bioactive molecules termed adipokines. One highly conserved family of adipokines is adiponectin/ACRP30 and its structural and functional paralogs, the C1q/tumor necrosis factor-alpha-related proteins (CTRPs) 1-7. Unlike adiponectin, which is expressed exclusively by differentiated adipocytes, the CTRPs are expressed in a wide variety of tissues. These proteins are thought to act mainly on liver and muscle tissue to control glucose and lipid metabolism. An analysis of the crystal structure of adiponectin revealed a structural and evolutionary link between TNF and C1q-containing proteins, suggesting that these proteins arose from a common ancestral innate immunity gene. Multiple isoforms of mouse CTRP4 have been reported.

Of the CTRPs, CTRP2 is most similar structurally and functionally to adiponectin. Recombinant CTRP2 rapidly activated AMPK and MAPK in cultured C2C12 cells, leading to increased glycogen accumulation and fatty acid oxidation.
