Description: The tumor necrosis factor (TNF) and TNF receptor (TNFR) gene superfamilies regulate numerous biological functions including cell proliferation, differentiation, and survival through regulating the activation of the transcription factor NF-kB and various mitogen-activated protein kinases. Nerve growth factor receptor (NGFR) was one of the earliest characterized members of this family. Also known as the low-affinity receptor p75NTR, this receptor is involved in several diverse functions such as apoptosis, neurite outgrowth during development, and myelination. Its ligands include NGF, brain-derived neurotrophic factor (BDNF), NT3, and NT4. NGFR can also associate with other NGF receptors such as Trk through the cytosolic and transmembrane domains and thus can function as a co-receptor that refines Trk affinity and specificity for neurotrophins. Finally, upon binding of various neurotrophins, NGFR associates with tumor necrosis factor receptor-6 (TRAF6), suggesting that it can potentially function as a signal transducer for NGF signals through NGFR.

Description: Ankyrin (ANK) repeats mediate protein-protein interactions in diverse families of proteins. The number of ANK repeats in a protein can range from 2 to over 20. ANK repeats may occur in combinations with other types of domains. The structural repeat unit contains two anti-parallel helices and a beta-hairpin, with repeats stacked in a superhelical arrangement. LIAR, also known as ANKRD54, is a recently identified ANK repeat-containing protein that is predominantly expressed in tissues rich in cilliated cells, such as olfactory sensory neurons and is predicted to be important to cilia. At least three isoforms of LIAR are known to exist.

Description: The leucine-rich, glioma inactivated gene 1 (LGI1) was first identified as a candidate tumor suppressor gene for glioma and may play a role in other cancers. LGI1 is a member of a family of highly related proteins containing leucine-rich repeats (LRRs) which are highly similar to other transmembrane signaling molecules and receptors. LGI1 serves as a ligand to ADAM22, a metalloprotease localized at the synapse. Mutations in LGI1 account for nearly half of autodominant lateral temporal epilepsy (ADTLE), an epileptic syndrome characterized by focal seizures with predominant auditory symptoms. Two isoforms of LGI1 are known to exist; this LGI1 antibody will recognize only the longer form. Anti-LGI1 is ideal for research in Neuroscience and Cancer.

Description: Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Histone proteins are highly post-translationally modified with Histone H3 being the most extensively modified of the five histones. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Histone H3 Antibody is ideal for investigators involved in Cell Signaling, Epigenetics, Nuclear Signaling research and Signal Transduction research.

Description: Rockland produces a wide range of human GST antibodies in our laboratories. Select appropriate GST antibodies for your research by isotype, epitope, applications and species reactivity. There are 22 members of the human GST family of proteins. GST is responsible for the conjugation of reduced glutathione to a wide number of exogenous and endogenous hydrophobic electrophiles. The amino acid sequence GST is highly conserved in most organisms including mammals. GSTs proteins are typically homodimeric, with both heterologous GST dimers have been observed. GST monomers have an average molecular weight of approximately 25-28 kDa in size. Note a different form of non-human GST (Glutathione-S-Transferase) is used as a protein expression tag commonly in molecular biology applications. All anti-GST antibodies my not react with recombinant GST-fusion proteins.

Description: Rabbit Anti-VhH antibody can be used to detect single domain VhH antibodies. The single-domain antibody (sdAb) is a small (12 - 14 kDa) antibody fragment that consists of a monomeric variable domain derived from the heavy chain, also called a VhH antibody. These heavy chain only Fab-like domains have activity that is similar to a whole antibody, and they are able to bind to a specific antigens. SdAb's are derived from camelid species that include llamas, alpacas and camels. Camelids produce both classical (containing heavy and light chain fragments) and non-classical antibody structures (containing only a heavy chain). VhH antibodies are the smallest functional antigen-binding fragment that occurs in nature and these are now being used in biotechnology as a novel antibody scaffold. The small size of the VhH single domain antibody makes it very attractive for use in diagnostic imaging and they have potential for therapeutic activity.

Description: MEK1 antibodies detect the MEK1 isoform.  Mitogen-activated protein kinase kinase 1, also known as MKK or MEK1, is an integral component of the MAP kinase cascade that regulates cell growth and differentiation.   This pathway also plays a key role in synaptic plasticity in the brain.  Activated MEK 1 acts as a dual specificity kinase phosphorylating both a threonine and a tyrosine residue on MAP kinase.   MEK1 and MEK2 are about 80% identical to each other, and nearly identical within the kinase domain.   The MEK1 antibody is ideal for investigators involved in Neuroscience, Cell Signaling and Cancer Research.

Description: Aldh5A1 is a member of the aldehyde dehydrogenase superfamily, a group of NAD(P)(+)-dependent enzymes that catalyze the oxidation of a wide spectrum of aliphatic and aromatic aldehydes. Aldehyde dehydrogenase enzymes are thought to play a major role in the detoxification of aldehydes generated by alcohol metabolism and lipid peroxidation. Aldh5A1 is a mitochondrial NAD(+)-dependent succinic semialdehyde dehydrogenase. A deficiency of this enzyme, known as 4-hydroxybutyricaciduria, results in a disorder of the neurotransmitter 4-aminobutyric acid (GABA). Symptoms usually include static encephalopathy, associated with developmental delays, hypotonia, ataxia, speech defects, and seizures. At least two isoforms of Aldh5A1 are known to exist.

Description: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. Beclin-1, a principal regulator of autophagosome formation, is in turn regulated by Ambra1. Ambra1 associates with Beclin-1 through a region near its center as determined by yeast two-hybrid assay. Null mutations in this gene in mice resulted in embryonic lethality with severe neural tube defects associated with autophagy impairment, accumulation of ubiquitinated proteins, unbalanced cell proliferation and excessive apoptotic death. Furthermore, down-regulation of Ambra1 in cultured cells though RNA interference decreased the level of rapamycin- and nutrient starvation-induced autophagy. Multiple isoforms of Ambra1 are known to exist.
