Description: Interleukin-31 (IL-31) is a recently discovered T-cell cytokine closely related to IL-6 type cytokines and is preferentially produced by T helper type 2 cells. IL-31 activity is mediated through the ligand-induced oligomerization of a dimeric receptor complex containing IL-31 receptor A and oncostatin M receptor. In response to IL-31 binding, these proteins activate the JAK/STAT and the AKT signaling pathways. RNA levels of IL-31 receptor A and oncostatin M receptor are induced in activated monocytes but are expressed constitutively in epithelial cells. IL-31, when overexpressed in transgenic mice, results in the development of pruritis, alopecia, and skin lesions and in humans may result in atopic dermatitis, suggesting that IL-31 may represent a novel target for antipruritic drug development.

Description: HSF1, or heat shock factor 1, belongs to a family of Heat Shock transcription factors that activate the transcription of genes encoding products required for protein folding, processing, targeting, degradation, and function (2). The up-regulation of HSP (heat shock proteins) expression by stressors is achieved at the level of transcription through a heat shock element (HSE) and a transcription factor (HSF) (3, 4, 5). Most HSFs have highly conserved amino acid sequences. On all HSFs there is a DNA binding domain at the N-terminus. Hydrophobic repeats located adjacent to this binding domain are essential for the formation of active trimers. Towards the C-terminal region another short hydrophobic repeat exists, and is thought to be necessary for suppression of trimerization (6). There are two main heat shock factors, 1 and 2. Mouse HSF1 exists as two isoforms, however in higher eukaryotes HSF1 is found in a diffuse cytoplasmic and nuclear distribution in un-stressed cells. Once exposed to a multitude of stressors, it localizes to discrete nuclear granules within seconds. As it recovers from stress, HSF1 dissipates from these granules to a diffuse nuceloplasmic distribution. HSF2 on the other hand is similar to mouse HSF1, as it exists as two isoforms, the alpha form being more transcriptionally active than the smaller beta form (7, 8). Various experiments have suggested that HFS2 may have roles in differentiation and development (9, 10, 11). Description: Interleukin-34 (IL-34) was identified through a functional screening of a library of secreted proteins after transfection of an embryonic kidney cell line with recombinant cDNAs. Flow cytometry and monocyte viability assays demonstrated increased survival and/or proliferation of monocytes induced by IL-34 and specific binding of IL-34 to CD14+ monocytes. IL-34 is a second ligand to the colony stimulating factor (CSF)-1 receptor in addition to CSF1. Like CSF1, IL-34 stimulates phosphorylation of ERK1 and ERK2 in human monocytes and induces differentiation of bone marrow cells into macrophages.

Description: IL-36G is a member of the interleukin 1 cytokine family whose gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2 (1). IL-36G is thought to activate the NF-kappaB pathway through IL-1 receptor family members IL-1RL2 and IL-1RAcP (2). Like the related proteins IL-36A and IL-36B, IL-36G requires post-translational processing for full agonist activity, but the cleavage mechanism is currently unknown (3). IL-36G expression is stimulated by interferon-gamma, tumor necrosis factor-alpha and interleukin 1 (4). The IL-36 cytokines have been suggested to amplify Th1 responses by enhancing proliferation and Th1 polarization of naive CD4+ T cells (5).

Description: The GDNF-inducible zinc finger protein 1 (GZF1) is a sequence-specific transcriptional repressor with a BTB/POZ domain and ten zinc finger motifs whose expression is required for renal branching morphogenesis during kidney development. GZF1 binds to the 5'regulatory region of the homeodomain protein HOXA10, suggesting that GZF1 may play a role in morphogenesis other than kidney development. Recent experiments have indicated that GZF1 associates with nucleolin and this association is mediated by the first four zinc finger motifs of GZF1. It is thought that Nucleolin modulates the subcellular localization of GZF1 as well as its transcriptional repressor activity.

Description: The HS-1 associated protein X-1 (Hax1) was initially identified in a yeast two-hybrid assay on the basis of its ability to bind to the hemapoietic cell-specific protein 1 (HS-1). Hax1 possesses anti-apoptotic activity and is structurally related to Bcl-2 family members, including the presence of BH1- and BH2-like domains. It has recently been shown to interact with HIV viral protein R (Vpr), a protein required for viral pathogenesis of HIV and linked to T-cell apoptosis through activation of caspases 3 and 9. Other studies indicate that Hax1-mediated processing of HtrA2 (also known as Omi) by the mitochondrial protease PARL allows survival of lymphocytes and neurons when cytokines are limiting. At least four isoforms of Hax1 are known to exist. This antibody is expected to recognize the longest isoform (Hax1a) as well as the shortest.
