Enzymes

This enzyme is a uracil N-glycosylase (UNG) obtained from the bacterium Esherichia coli. UNG enzymes specifically hydrolyze the N-glycosidic bond of uracil residues in ss/dsDNA molecules leaving an abasic site in the strand but will not react with RNA. The optimal temperature range for E. coli UNG activity is from 30 to 40 °C. The enzyme is irreversibly inactivated after ten minutes of incubation at temperatures ≥95 °C making it suited for carryover prevention in standard PCR and qPCR applications. It is active in most standard reaction buffers used for derivatives of Taq and Pfu DNA polymerases.
