Polymyxin B sulfate is a mixture of Polymyxin B1 and B2 sulfate. It is used clinically to treat infections of the urinary tract, meninges, and blood stream, caused by susceptible strains of Pseudomonas aeruginosa. It is used to study multidrug-resistant pathogens.
Mode of Action: Polymyxin B Sulfate binds to the lipid A portion of bacterial lipopolysaccharides†, disrupting the cytoplasmic membrane by inducing pores large enough to permit nucleotide leakage in bacterial walls. This disrupts the permeability of the cytoplasmic membrane.
Polymyxin B inhibits gram-negative organisms in vitro wherein it binds to and interferes with the permeability of the bacterial cytoplasmic membrane and is also reported to inhibit Ca2+ dependent protein kinase C (PKC).
