Description: Glutamate receptors play an important role in neural plasticity, development and degeneration. The glutamate receptor interacting proteins, GRIP1 and GRIP2, members of the PDZ domain-containing protein family, mediate the trafficking and membrane organization of a number of transmembrane proteins. GRIP1 and GRIP2 specifically bind to the AMPA receptor subunits, GluR 2/3 and are involved in the targeting of GluR 2/3 to the synapse. GRIP1 is expressed in early development before the expression of AMPA receptors, while GRIP2 expression parallels that of AMPA receptors during later developmental stages. GRIP1 and GRIP2 may be involved in the induction of cerebellar long-term depression (LTD).

Description: The human trans-activation response (TAR) RNA binding protein 1 (TRBP1) was initially identified as a protein that binds to the HIV-1 TAR RNA and activates the long terminal repeat (LTR) expression in the absence and presence of the viral trans-activator Tat. This binding is enhanced by the presence of co-factors such as elongation factor 1a (EF-1a), polypyrimidine tract-binding protein (PTB), and the chaperonin-like protein stimulator of TAR RNA-binding proteins (SRB). TRBP1 may act to disengage RNA polymerase II from TAR during transcriptional elongation if the RNA polymerase stalls at the TAR during transcriptional elongation.

Description: The ZBTB family of proteins is comprised of diverse zinc finger proteins that also contain a BTB (BR-C, ttk and bab) domain. While little is known about ZBTB3, the related protein ZBTB2 is thought to be phosphorylated in response to the DNA damage, probably by either ATM or ATR. Other ZBTB proteins, such as ZBTB4 and ZBTB38 bind methylated DNA and repress transcription, suggesting that ZBTB3 may also act as a transcription repressor.

Description: WDR61 (WD-repeat-containing protein 61), also known as SKI8 or REC14, is a subunit of the human PAF and SKI complexes, which function in transcriptional regulation and are involved in events downstream of RNA synthesis, such as RNA surveillance (1,2). PAF1C is implicated in regulation of development and maintenance of embryonic stem cell pluripotency. Component of the SKI complex which is thought to be involved in exosome-mediated RNA decay and associates with transcriptionally active genes in a manner dependent on PAF1C (3,4).

Description: NALP proteins include the apoptosis regulator APAF1 (apoptotic protease activating factor 1) and mammalian NOD-LRR proteins and are thought to be involved in inflammation and reproduction. NALP6, also known as PYPAF5, is highly expressed in granulocytes and T cells and co-localizes with the PYRIN-CARD protein ASC. Co-expression of NALP6 with ASC results in a synergistic activation of NF-kB and activated caspase-1-dependent cytokine processing, suggesting that NALP6 functions in immune cells to coordinate the transduction of pro-inflammatory signals.

Description: Nicalin, along with its binding partner Nomo, is a novel signaling antagonist to Nodals, signaling factors of the TGF-b superfamily that play a key role in vertebrate development. Nicalin is distantly related to Nicastrin, a component of the Alzheimer's disease-associated gamma-secretase, and is thought to have a similar function. Another protein, TMEM147, has also been recently identified as associating with Nicalin and NOMO as part of the gamma-secretase-like complex.

Description: The NEDD4 family-interacting protein 1 (NDFIP1) belongs to a small group of evolutionarily conserved proteins with three transmembrane domains and is an integral Golgi membrane protein. It is a potential target for ubiquitination by the Nedd4 family of proteins. NDFIP1 is strongly expressed in surviving neurons following acute cortical brain injury, and overexpression in cultured cortical neurons increased survival following growth factor starvation, suggesting that NDFIP1 may play a role in neuronal survival. NDFIP1 and the related protein NDFIP2 are thought to interact with and regulate multiple components of the EGF and PTEN/Akt signaling pathways. Recent studies suggest that NDFIP1 may also play a role in Th17 differentiation by limiting the production of proinflammatory cytokines.

Description: IL-36A is a member of the interleukin 1 cytokine family whose gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2 (1). IL-36A is thought to activate the NF-kappaB pathway through IL-1 receptor family members IL-1RL2 and IL-1RAcP (2). Like the related proteins IL-36B and IL-36G, IL-36A requires post-translational processing for full agonist activity, but the cleavage mechanism is currently unknown (3). The IL-36 cytokines have been suggested to amplify Th1 responses by enhancing proliferation and Th1 polarization of naive CD4+ T cells (4).

Description: The interleukin 17 receptor E (IL-17RE) shares limited similarity to the receptor of IL-17A, a ubiquitous type I membrane glycoprotein that with IL-17A plays a pathogenic role in many inflammatory and autoimmune diseases such as rheumatoid arthritis. IL-17RE is the functional receptor of IL-17C and has been suggested to mediate mucosal immunity to infection with intestinal pathogens. IL-17C is produced by epithelia in response to bacterial challenge and binds to a receptor complex formed by IL-17RE and IL-17RA on the epithelial cell surface, thereby regulating the immune function of epithelial cells in an autocrine manner.

Description: Interleukin-31 (IL-31) is a recently discovered T-cell cytokine closely related to IL-6 type cytokines and is preferentially produced by T helper type 2 cells. IL-31 activity is mediated through the ligand-induced oligomerization of a dimeric receptor complex containing IL-31 receptor A and oncostatin M receptor. In response to IL-31 binding, these proteins activate the JAK/STAT and the AKT signaling pathways. RNA levels of IL-31 receptor A and oncostatin M receptor are induced in activated monocytes but are expressed constitutively in epithelial cells. IL-31, when overexpressed in transgenic mice, results in the development of pruritis, alopecia, and skin lesions and in humans may result in atopic dermatitis, suggesting that IL-31 may represent a novel target for antipruritic drug development.

Description: The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Other forms of CD45 with restricted cellular expression include CD45RA, CD45RB, CD45RO and several others. The RA3-6B2 antibody is one of the most consistently used leukocyte markers for B cells, T cell subsets and NK cell subsets in human and mouse. This antibody has also been reported as cross-reactive with feline CD45R/B220.

Description: Glial cell line-derived neurotrophic factor (GDNF) plays key roles in the control of vertebrate neuron survival and differentiation. A novel neurotrophic factor was recently cloned from human and mouse and designated neurturin. Physiological responses to neurturin (NTN) require the presence of receptor tyrosine kinase RET and a novel glycosylphosphatidylinositol linked receptor NTNRalpha. The cDNAs encoding NTNRalpha from human, rat, chicken, and mouse have been cloned recently and termed GDNFRb, Ret ligand 2 (RETL2) or TGF-beta-related neurotrophic factor receptor 2 (TrnR2) and nominated as GFRa-2 recently. NTN binds to and forms a complex with GFRa-2 and the Ret PTK and activates the RET receptor tyrosine kinase pathway. Both NTN and GDNF can activate the MAP kinase and phosphatidylinositol 3-kinase pathways and play a critical role in the development of many neuronal populations. Neurturin and GDNF define a new family of neurotrophic factors.

Description: OCC-1 (Overexpressed in colon carcinoma-1) was initially identified as a novel human cDNA overexpressed in a colon carcinoma cell line. Elevated levels of OCC-1 mRNA were observed in three of eight colon carcinomas, suggesting that OCC-1 may be a hallmark of only a subset of colon carcinomas. It is highly expressed in skeletal muscle, kidney, placenta, and pancreas tissues, with low to no expression in heart, brain, lung and liver tissues. At least two different OCC-1 mRNA species are known to exist.
