Lysosomal Associated Membrane Protein 2 (LAMP2), also known as CD107b and Mac-3, is a glycoprotein found in the lysosomal membrane that plays a crucial role in cellular processes such as chaperone-mediated autophagy. LAMP2 is encoded by the LAMP2 gene, which can produce three isoforms: LAMP-2A, LAMP-2B, and LAMP-2C, each with distinct functions. LAMP-2A is involved in transporting proteins into lysosomes for degradation, LAMP-2B is essential for the fusion of autophagic vacuoles with lysosomes, and LAMP-2C is linked to the breakdown of nucleotides. Mutations in LAMP2 can lead to Danon disease, characterized by cardiomyopathy, myopathy, and intellectual disability, and have been associated with increased susceptibility to oxidative stress and ferroptosis. LAMP2 is also implicated in cancer metastasis, where it mediates tumor cell adhesion and invasion.
